Histochemical Study of Cutaneous Mucins in Hypothyroid Dogs

Abstract: Abstract. A dermal mucinoisis, visualized as dermal alcianophilic material, is occasionally present in canine hypothyroidism (myxedema). Various histochemical reactions (alcian blue/periodic acid-Schiff [PAS], alcian blue at pH 2.6, alcian blue at pH 1.0, critical electrolytical concentrations with and without dimethylsulfoxide, differential hydrolysis by hyaluronidases) were performed on skin biopsies from six dogs (four females and two males ranging from 8 to 13 years) affected by hypothyroidism, all of them… Show more

“…Positive alcian blue staining indicates the presence of acid mucus polysaccharides (Doliger et al, 1995; Sasai, 1971). To further characterize the swollen connective tissues of bulgy morph tadpoles, sections were treated with 1% alcian blue 8GS-1N HCl (pH 1.0, for strongly sulfated glycosaminoglycans) or 1% alcian blue 8GS-3% acetic acid (pH 2.5, for a broad range of acidic glycosaminoglycans).…”

“…In acromegaly the skin thickening has been ascribed to mucopolysaccharide accumulation (Matsuoka et al 1982). Similarly, in dogs with primary hypothyroidism the thickening of the skin is also due to mucopolysaccharide accumulation (Muller et al 1989, Doliger et al 1995. Thus, some of the physical changes observed in dogs with primary hypothyroidism might very well be acromegalic changes.…”

“…One histochemical investigation 5 demonstrated that the mucinous material deposited in the skin of hypothyroid dogs was Alcian blue (AB)/Schiff's periodic acid (PAS)‐positive corresponding to acid glycosaminoglycans (GAGs), concluding that probably the only polysaccharide compound involved in canine dermal mucinosis was hyaluronic acid (HA). Another study 6 demonstrated that in the dermis of shar‐pei dogs with cutaneous mucinosis, the predominant mast cells were chymase‐positive, instead of the more common tryptase‐positive, suggesting that these cells might play a role in the pathogenesis of this disease.…”
