“…Short T2* water fraction parallels changes are not considered (18). In addition, the Mankin grading system includes as a criterion safranin O staining intensity, which is affected by protocol variability, such as tissue fixation and decalcification, and these factors could lead to errors in the measure (39,40). Fourth, there is a discrepancy in the image scales between histologic sections that are a few micrometers in thickness and MR images that are several millimeters thick.…”

“…Thus, the Safranin O-Fast Green staining protocol has been widely used as a marker for PG loss. 28 After staining, the AC slices were rinsed in 95% ethanol and mounted in slides. All histological sections were automatically imaged by a bright-field digital slide scanner with a 20× dry objective (ScanScope AT, Aperio, Vista, CA).…”

“…The OCT embedding material was removed from the FTIRI samples by adding a few drops of de-ionized water and rapid removal with a pipette prior to air-drying the sample. Histological samples were fixed in neutral buffered formalin and stained with Safranin-O (Hyllested et al, 2002) and all specimens were evaluated histologically for OA severity.…”
