Histochemical Methods for Separate, Consecutive and Simultaneous Demonstration of Acetylcholinesterase and Norepinephrine in Cryostat Sections

Abstract: Techniques for the separate, consecutive and simultaneous demonstration of acetylcholinesterase and norepinephrine in cryostat sections are described and discussed. The results, obtained in a variety of tissues from different animal species, are as satisfactory—in terms of sensitivity and sharpness of localization—as those obtained by previously described methods. The simultaneous visualization of both acetylcholinesterase and norepinephrine in the same section provides hitherto unavailable data relative to th… Show more

“…The chickens and pheasants were killed by aether narcosis and their bursae cloacales were removed. ACHE-positive nerve components were visualized by the direct tiocholin method cytochemistry evidence acetylcholinesterase by El-Badawi and Schenk (1967), adrenergic nerve components we visualized with glyoxylic acid in modification of Shvalev and Zhuckova (1987). Microscopic examination and photo documentation were performed using the NIKON.…”

“…• C. The acetylcholinesterase (AChE)-positive fibers in the thymus and bursa of Fabricius were visualized by a direct thiocholine method according to El Badawi & Schenk (1967) to provide cytochemical proof of AChE. In this method acetylcholinesterase present in the cholinergic nerves releases thiocholine from acetylthiocholine, which reduces potassium ferricyanide to potassium ferrocyanide, capable of producing insoluble copper ferrocyanide with copper ions (Hatchett's brown).…”

“…The specific fluorescence of adrenergic nerve fibres was studied in incident ultraviolet light (epiluminiscence) using a microscope Jenalumar 2 (Zeiss, Jena). The presence of acetylcholinesterase in splenic neural structures was determined by the direct thiocholine method modified by El-Badawi and Schenk (1967). After incubation, the individual sections were mounted on slides and examined under a light microscope Jenalumar 2.…”
