Histochemical Demonstration of Two Subtypes of O‐linked Oligosaccharides in the Rat Gastric Glands

Abstract: The gastric glands synthesize glycoproteins whose oligosaccharides are linked to the peptide core mainly by the O-glycosidic bond, specifically removed by beta-elimination procedure. Our aim was to research the possibility of the existence of two subtypes of O-linked oligosaccharides with a different behavior to the removal procedure. The lectins from peanut (PNA) and Maackia amurensis (MAA-I) were histochemically used as markers of the O-linked oligosaccharides. Sections were also pretreated with beta-elimina… Show more

“…This could be explained by the unmasking of GalNAc moieties in N ‐glycans, which are initially inaccessible to the lectin, and the removal of O ‐glycans allows the labeling by DBA. The unmasking of carbohydrates by deglycosylation techniques has been described previously (Alonso et al,2001; Sáez et al,2001; Gómez‐Santos et al,2007).…”

“…To remove N‐linked glycans, incubation with 40 U/mL recombinant PNGase F in PBS at 37°C for 72 h was performed as previously described (Sáez et al,2000a; Gómez‐Santos et al,2007). As control for the complete N ‐glycan removal, rat testis sections were stained with Galanthus nivalis agglutinin (GNA, EY Laboratories, 60 μg/mL working dilution).…”

“…However, a reduction of labelling intensity after each pretreatment is to be expected, which is not evident with some lectins in our work. These amazing results are also common, and have been explained by assuming that each deglycosylation method unmasks some glycans which were previously inaccessible to the lectin (Alonso et al 2001;Gómez-Santos et al 2007;Sáez et al 2001b;Desantis et al 2006;Parillo et al 2009). Another possible explanation is that lectins could be labelling not only glycoproteins, but also glycolipids.…”

“…β‐elimination was carried out following the technique previously described (Sáz et al,2000a) for five, seven, and twelve days, (Valbuena et al,2010). To remove N‐linked glycans, incubation with recombinant PNGase F was performed as previously described (Gómez‐Santos et al,2007; Sáez et al,2000a).…”
