Hippocampal mossy fiber long‐term depression in Grm2/3 double knockout mice

Abstract: Group II metabotropic glutamate receptors (mGluR2, encoded by Grm2, and mGluR3, encoded by Grm3) are inhibitory autoreceptors that negatively modulate the adenylate cyclase signaling cascade. Within the hippocampus, mGluR2 is believed to play a key role in the induction of long-term depression (LTD) at mossy fiber-CA3 synapses. Here, we used Grm2/3 double knockout (dko) mice to investigate to what extent group II mGluRs are necessary for mossy fiber LTD. Surprisingly, we found that these mice displayed promine… Show more

“…It is plausible that the reduced dopamine ‘tone’ reported here contributes to these findings. Whether it also underlies the arousal-dependent effect on spatial memory [10] or the impaired long-term depression [22] of the mice, is unknown. But it may be significant that the NAc is involved in motivation, reward and aversion [23,24].…”

“…Brains were taken from adult Grm3 −/− knockout mice, Grm2 −/− /3 −/− double knockout mice, and wild-type mice (Lyon et al, 2008, Lyon et al, 2011a, De Filippis et al, 2015), snap frozen, and stored at − 80 °C. For protein isolation, a small piece of frontal tissue was cut with a clean razor blade, weighed, and homogenised using membrane extraction buffer and a Dounce homogeniser on ice ( n  = 3 adult female mice per genotype).…”

“…Unexpectedly, two recent studies have called into question the induction mechanism underlying MF-LTD. Using more specific and potent group II mGluRs antagonists, as well as mGluR2/3 double knockout mice, these studies still observed presynaptic MF-LTD (Wostrack and Dietrich 2009;Lyon et al 2011). MF-LTD induced in mGluR2/3 double knockout mice was abolished by pharmacologically blocking kainate receptors (Lyon et al 2011), raising the intriguing possibility that these receptors, via a metabotropic action, could be involved in the induction of presynaptic MF-LTD (Negrete-Diaz et al 2007).…”
