Highly Tumorigenic Diffuse Large B Cell Lymphoma Cells Are Produced by Coculture with Stromal Cells

Abstract: Background/Aims: Diffuse large B cell lymphoma (DLBCL) is heterogeneous. We aimed to explore how tumor microenvironment promotes lymphoma cell aggressiveness and heterogeneity. Methods: We created a coculture system using human DLBCL cells and mouse bone marrow stromal cells. Proliferative capacity, drug resistance, clonogenicity, and tumorigenicity were compared in lymphoma cells from the coculture system and lymphoma cells cultured alone. Expression of Notch signaling associated genes was evaluated using rea… Show more

“…22,23 Therefore, discovering a novel reagent or drug targeting LSCs might hold promise for the clinical treatment of AML. According to former studies, 24,25 Traditional Chinese medicine (TCM) normally demonstrates drug acceptance when applied in the disease therapy. Therefore, we speculated that TCM might play a feasible role in treating the senescence of CD34 + CD38 − LSCs in certain signaling pathways.…”

“…In DLBCL, the upregulation of Notch-3 in tumor cells under close cell-cell contact with BM-derived stromal cells has been implicated in the development of aggressive lymphoma cells ( 61 ). In turn, such direct interaction between DLBCL cells and stromal cells mediates an increase in B-cell activating factor (BAFF) expression by stromal thus resulting in a decrease of chemotherapy-induced B-cell apoptosis ( 62 , 63 ) ( Table 1 ).…”

“…The role of the BM stromal cells is also prominent in DLBCL. A recent study reported that co-culture of DLBCL cells with stromal cells leads to highly tumorigenic cells ( Lin et al, 2018 ). The mechanism by which the BM stromal cells could induce DLBCL cell growth may be mediated by secreting IL-6 and in a JAK2/STAT3 dependent manner shown in both in vivo and in vitro systems.…”
