Highly specific and rapid immuno-fluorescent visualization and detection of E. coli O104:H4 with protein-A coated magnetic beads based LST-MUG assay

Abstract: A method combining immunomagnetic separation and fluorescent sensing was developed to detect Escherichia coli (E. coli) O104:H4. The antibody specific to E. coli O104:H4 was immobilized on protein A-coated magnetic beads. This protein-A-anti E. coli O104:H4 complex was used to bind Fluorescein IsoThioCyanate (FITC) labeled E. coli O104:H4 antigen (whole cell) on it. The goal was to achieve a fluorescently detectable protein-A-anti E. coli O104:H4-E. coli O104:H4 complex on the magnetic beads. Fluorescent micro… Show more

“…The proposed system is comparable with or even better than other enzyme-based assays. [33][34][35][36][37][38] More importantly, peptide functionalized magnetic beads offer the benefits of simplicity, speed, stable and flexibility to perform potentiometric detection based on surface charge change. The detection time is shorter than most commercially available detection kits for E. coli.…”

“…The protein A-coated magnetic beads can be used to immuno-precipitate target proteins from crude cell lysates using selected primary Abs. In addition, specific Abs can be chemically cross-linked to the protein A-coated surface to create reusable immuno-precipitation beads, thus avoiding the co-elution of antibodies with target antigens [37]. The streptavidin-coated magnetic beads (Strept-MBs) provide a fast and convenient method for manual or automated immuno-precipitation, protein interaction studies, DNA-protein pull downs and the purification of biotinylated proteins and nucleic acids.…”
