Highly conserved glucose-regulated protein in hamster and chicken cells: preliminary characterization of its cDNA clone.

Abstract: A temperature-sensitive mutant K12 derived from a Chinese hamster fibroblast has been shown to overproduce three specific proteins of Mr 94,000, 78,000, and 58,000 when incubated at the nonpermissive temperature (40.50C). We previously identified these proteins as glucose-regulated proteins similar to those observed in chicken embryo fibroblasts when the cells are starved of glucose. In this report, we show that the Mr 78,000 proteins isolated from the hamster K12 cell line and from chicken embryo fibroblasts… Show more

“…On the amino acid level, overall identity with PDI is 33 %, and identity with the a-b-bh-ah domains of ERp72 is 40 %. The protein responds to stress stimuli in a manner similar to that of the glucose-regulated proteins GRp78 and GRp94 [146,147], and may be up-regulated additionally by oestrogens in brain tissue and by luteinizing-hormone-releasing hormone in the pituitary [148].…”

“…The Chinese hamster fibroblast cell lines WglA and K12 have been described previously (9,17). K12 GRP94, respectively, has been described previously (11,12). The pSVOCAT plasmid was provided by B. Howard.…”

“…Using the hamster mutant cell line K12, which overproduces these GRPs at the nonpermissive temperature, we previously isolated two cDNA clones, p4A3 and p3C5, which by the criterion of hybrid select translation encode the hamster GRPs of 94 and 78 kilodaltons (GRP94 and GRP78), respectively (11,12). These cDNAs are encoded by singlecopy genes whose transcription is highly inducible by glucose starvation in a variety of animal cells (13).…”

“…This ERlocalized hsp70 protein, initially described as the mammalian glucose-regulated protein grp78 (Lee et al, 1981 : Munro and Pelham, 1986) is induced by the accumulation of misfolded proteins inside the ER (Kozutsumi et al, 1988;Normington et al, 1989: Rose et al, 1989. Early studies showed that BiP binds to unassembled immunoglobulin heavy chains (Copeland et al, 1986;Gething et al, 1986: Haas andWabl, 1983).…”
