High-Titer Lactic Acid Production by Pediococcus acidilactici PA204 from Corn Stover through Fed-Batch Simultaneous Saccharification and Fermentation

Abstract: Lignocellulose comprised of cellulose and hemicellulose is one of the most abundant renewable feedstocks. Lactic acid bacteria have the ability to ferment sugar derived from lignocellulose. In this study, Pediococcus acidilactici PA204 is a lactic acid bacterium with a high tolerance of temperature and high-efficiency utilization of xylose. We developed a fed-batch simultaneous saccharification and fermentation (SSF) process at 37 °C (pH 6.0) using the 30 FPU (filter paper units)/g cellulase and 20 g/L corn st… Show more

“…The untreated corn stover (% w/w, on a dry basis) contained mainly cellulose (31.18 ± 0.23), hemicellulose (26.95 ± 0.14), and lignin (16.43 ± 0.27). The CS used in this study contained a lower amount of cellulose, while amounts of hemicellulose and lignin were comparable to those of reported works [5,10]. It was likely that differences in the components of CS might be due to its trait variations and growing lands.…”

“…The inhibitors derived from the pretreatment of CS have adverse impact on sacchari cation by cellulase. The effect could be eliminated via the operation of washing, biodetoxi cation, sodium bisul te addition, etc [7,10,17]. However, washing would generate a large quantity of waste water while biodetoxi cation need a long period.…”

“…The values of OD 600 can represent the dynamic growth rates of the microbial cells [ 31 , 32 ]. From Figure 1 a, it was found that the values of OD 600 at day 1 during fermentation with various media were appropriately 0.14 ± 0.01.…”

“…Many studies have been conducted to evaluate the degradation of lignocellulosic biomass and the production of LA using physical, chemical, and biological pretreatments. Dilute acid pretreatment was used among these pretreatment methods to efficiently produce lactic acid [ 7 , 9 , 10 ]. A high LA titer of 101.9 g/L was obtained from sulfuric acid pretreated and bio-detoxified corn stover (CS).…”
