High‐throughput logP measurement using parallel liquid chromatography/ultraviolet/mass spectrometry and sample‐pooling

Abstract: A novel approach to high-throughput logP measurement based on liquid chromatography/ultraviolet/mass spectrometry (LC/UV/MS) is proposed. The logP value is determined by correlation with the logk value, where k is the capacity factor k = (t(r)-t(0))/t(0), with the logP value using a defined set of standards. Since the analyte retention time (t(r)) is determined from the appropriate extracted ion chromatogram (EIC), there are no interferences from impurities and this allows the pooling of multiple compounds int… Show more

“…Shake-flask, microshake-flask, and chromatography retention are the most common methods for compound lipophilicity measurement and some of them could be used in a throughput mode [25][26][27][28][29][30][31]. Despite the availability of these assays, different instruments and multiple measurements are often needed to obtain LogP, LogD (pH) , and pK a values, separately.…”

“…Subsequent papers demonstrate the use of MUX technology in the quantitative analysis of samples derived from rat [182] human [183], mouse, rat, rabbit and dog [184,185] plasmas and the analysis of rat liver microsomal samples [186]. Eightchannel MUX systems have also been employed for the determination of logP [187] and to carry out accurate mass measurements [188].…”

“…Fluorescein sodium, which is freely soluble in water (0.1-1.0 g/mL) (36) and has a log P value of -1.52 (37), was chosen as a hydrophilic compound. Triamterene, a potassium-sparing diuretic with water solubility of 28 µg/mL (38) and log P value of 1.25 (39), was used as a fluorescent hydrophobic model compound. The fluorescent model substances were embedded in a blend of ammonio methacrylate copolymer type A and B (Eudragit RL/Eudragit RS, 3:7 ratio).…”
