High-Throughput Automated Microscopy of Circulating Tumor Cells

Abstract: Circulating tumor cells (CTCs) have the potential of becoming the gold standard marker for cancer diagnosis, prognosis and monitoring. However, current methods for its isolation and characterization suffer from equipment variability and human operator error that hinder its widespread use. Here we report the design and construction of a fully automated high-throughput fluorescence microscope that enables the imaging and classification of cancer cells that were labeled by immunostaining procedures. An excellent … Show more

“…In this study, cells categorized as CTCs were automatically classified by the algorithm implemented in the imaging system, these events were further corroborated by a trained operator. A detailed description and validation of the imaging system performance can be found in Aguilar-Avelar et al 45 . In addition, if molecular analysis are required, the membrane can be collected in a microtube for nucleic acid extraction using commercially available kits.…”

“…Up until now, most of the micro uidic chip-based CTCs platforms still rely on manual operations for cell screening and interpretation after immuno uorescent staining of target cells, which is time-consuming and causes inconsistent results. So far, only a limited number of automated CTCs microscope systems have been reported 21 . The automated CTCs scanning and identi cation system can complete the scanning of the target cells in a short time, which not only saves time and manpower, but also helps to establish standard processes.…”

“…The AccuCyte-CyteFinder was able to identify equivalent or greater numbers of CTCs found by the CellSearch system [ 68 ]. Aguilar-Avelar et al (2019) described the design and construction of a fully automated high-throughput fluorescence microscope that enables the recognition, imaging, and classification of CTCs in a blood sample that were labeled by immunostaining [ 69 ]. The microscope hardware accurately discriminated CTCs among cells present in blood and the hardware efficiently captured light emitted from unstained cells while the fluorescence signals were used to automatically classify and precisely identify CTCs on a sample by thousands of background cells [ 69 ].…”
