RESUMO.O feijão (Phaseolus vulgaris L.) é de grande expressão alimentícia. A emergência da cultura é dependente de água e considerada a fase mais crítica. Os objetivos deste trabalho foram simular a deficiência de água no início da germinação em laboratório, em sementes de feijão 'Pérola', utilizando-se: manitol, CaCl 2 , MgCl 2 e NaCl em potenciais de 0; -0,3; -0,6; -0,9 e -1,2 MPa, estabelecidos pela equação de Van't Hoff e avaliar o perfil eletroforético de proteínas totais solúveis por meio de SDS-PAGE. Foram avaliados: germinação, classificação de vigor, massa seca de raiz e de parte aérea e resposta diferencial de expressão de proteínas. O delineamento experimental foi inteiramente casualizado. Os dados foram analisados pela aplicação do teste F, para análise de variância, regressão polinomial para os níveis de potencial osmóticos para cada uma das variáveis fisiológicas estudadas. O bandeamento eletroforético foi avaliado visualmente por imagem dos géis. A simulação do estresse permitiu avaliar a drasticidade do NaCl em todos os parâmetros avaliados e a ausência de proteínas de baixo peso molecular neste osmótico. As proteínas de 110 e 30 kDa foram indicativas de estresse hídrico, mas não do salino.Palavras-chave: Phaseolus vulgaris L., deficiência de água, HSPs.ABSTRACT. Differential protein expression during germination as a result of a water deficit associated with variable osmotic pressure in snap-beans. Snap-beans (Phaseolus vulgaris L.) have a high nutritionally value. Successful seedling emergence is dependent on the availability of water and is considered the most critical phase in plant development. The objectives of this work were to simulate water deficiency at the beginning of the germination period. Snap-bean seeds of the variety 'Pérola' were submitted to osmotic stress under laboratory conditions. Mannitol, CaCl 2 , MgCl 2 and NaCl were used to develop five degrees of osmotic potential: 0, -0.3, -0.6, -0.9 and -1.2 MPa. Van't Hoff's equation was used to calculate the osmotic potential. The protein pattern of the total soluble proteins after electrophoresis was evaluated with SDS-PAGE. The response parameters were evaluated including the germination, the vigour classification, the root dry weight, the shoot dry weight and the resultant differential protein expression. The experimental design was completely randomised. Data were analysed using the F test (ANOVA), and polynomial regression was used for the osmotic potential levels for each parameter. The protein banding pattern was evaluated using gel imaging. The simulation of a water deficiency resulted in the perception of a stress and a subsequent response to NaCl in all of the parameters that were evaluated. The harsh effects of the NaCl were validated as the lack of the expression of low molecular weight proteins. Proteins with molecular weights of 110 and 30 kDa were indicative of water stress but not of salinity.

Small heat shock proteins (sHSPs) function as ATP-independent molecular chaperones, and although the production and function of sHSPs have often been described under heat stress, the expression and function of sHSPs in fundamental developmental processes, such as pollen and seed development, have also been confirmed. Seed germination involves the breaking of dormancy and the resumption of embryo growth that accompany global changes in transcription, translation, and metabolism. In many plants, germination is triggered simply by imbibition of water; however, different seeds require different conditions in addition to water. For small-seeded plants, like Arabidopsis (Arabidopsis thaliana), lettuce (Lactuca sativa), tomato (Solanum lycopersicum), and tobacco (Nicotiana tabacum), light is an important regulator of seed germination. The facts that sHSPs accumulate during seed development, sHSPs interact with various client proteins, and seed germination accompanies synthesis and/or activation of diverse proteins led us to investigate the role of sHSPs in seed germination, especially in the context of light dependence. In this study, we have built transgenic tobacco plants that ectopically express sHSP, and the effect was germination of the seeds in the dark. Administering heat shock to the seeds also resulted in the alleviation of light dependence during seed germination. Subcellular localization of ectopically expressed sHSP was mainly observed in the cytoplasm, whereas heat shock-induced sHSPs were transported to the nucleus. We hypothesize that ectopically expressed sHSPs in the cytoplasm led the status of cytoplasmic proteins involved in seed germination to function during germination without additional stimulus and that heat shock can be another signal that induces seed germination.

ABSTRACT-Heat shock proteins (HSPs) are the proteins which are present normally in the cell but their expression level increases under stress condition and are mainly divided into five groups, low molecular weight HSP (LMW HSP), HSP 60, HSP 70, HSP 90 and high molecular weight HSP (HMW HSP). All these classes of HSPs are highly conserved and ubiquitous in nature and hence serve as a good model for phylogenetic analysis. For the first time in this study,the sequence and structural analysis has been carried out to predict evolution of HSPs. The results obtained clearly show higher degree of sequence and structural conservation. HSP 60 and HSP 70 are highly conserved in terms of both sequence and structure alignment in comparison to HSP 90. The minimum amino acid identity that has been observed between all the homologous sequences is 32.46%, 38%, 23.60% for HSP 60, HSP 70 and HSP 90 respectively, indicating HSP 70 as the most conserved protein family followed by HSP 60 and HSP 90 family. The structural analysis of these proteins showed dominance of beta sheets in HSP70 and helices in HSP 90. The detailed analysis of all the HSP homologues revealed high conservation of glycine residues and ATP binding pockets. Thus this study has revealed that HSPs are highly structurally and functionally conserved proteins and warrants further detailed analysis at organism level.
