“…NIH-3T3 cells were seeded in a 96-well plate at a density of 1500 cells/well and incubated in DMEM plus 10% FBS (v/v) at 37 °C and 5% CO 2 for 24 h, followed by the replacement of the culture medium with 100 μL of the freshly prepared leaching solution. Cells were cultured in the leaching solutions for 1, 2, 3, and 7 days and CCK-8 assay was then conducted to determine cell growth by measuring the absorbance at 450 nm using a Plate Reader (SynergyH1/H1M) . The LIVE/DEAD cell viability assay was performed with cells cultured in the leaching solution of the PAAm-ELN 1.0 -TA 0.2 -CS 0.06 hydrogel by staining the live and dead cells at different time intervals (1, 2, 3, and 7 days), respectively. , The stained cells were subsequently imaged using an inverted fluorescence microscope with excitation/emission at 495/515 and 495/635 nm for live and dead cells, respectively.…”
