High‐resolution 1H NMR and magic angle spinning NMR spectroscopic investigation of the biochemical effects of 2‐bromoethanamine in intact renal and hepatic tissue

Abstract: The metabolic consequences of xenobiotic-induced toxicity were investigated using high-resolution magic angle spinning (MAS) NMR spectroscopy of intact tissue. Renal papillary necrosis (RPN) was induced in Sprague-Dawley rats (n ‫؍‬ 12) via a single i.p. dose of 250 mg/kg 2-bromoethanamine (BEA) hydrobromide. At 2, 4, 6, and 24 h after treatment with BEA, three animals were killed and tissue samples were obtained from liver, renal cortex, and renal medulla. Tissue samples were also removed at 2 and 24 h from m… Show more

“…[28][29][30] It was performed in this study to assess the biological impact of SWCNT injection on liver metabolism as a general index of toxicity. HR-MAS 1 H NMR spectroscopy of intact tissue enables the simultaneous measurement of both lipids and low molecular weight metabolites, which is important in a toxicological diagnosis.…”

“…The decrease in PUFA can be attributed to the increased stimulation of fatty acid oxidation which is another additional alternative pathway for energy production [ 101 when the oxidative mitochondrial pathway is inhibited by CyA. However, this adaptive process that results in lipid peroxidation is known to be harmful to the renal cortex and medulla [6]. Indeed, various metabolic markers for nephrotoxicity were found in rat kidney tissue treated with CyA.…”

“…Magic Angle Spinning NMR (MAS-NMR) involves the detection of intact tissues and cells with little or no preparation [53,54]. MAS-NMR significantly reduces the effect of magnetic field inhomogeneity in ordered materials.…”
