High resolution proteome/peptidome analysis of body fluids by capillary electrophoresis coupled with MS

Abstract: All organisms contain thousands of proteins and peptides in their body fluids. A deeper insight into the functional relevance of these polypeptides under different physiological and pathophysiological conditions and the discovery of specific peptide biomarkers would greatly enhance both diagnosis and therapy of specific diseases. Proteomic methods can provide means to accomplish this grand medical vision. In this review, we will focus on the potential use of proteome analysis for clinical applications, such as… Show more

“…Candesartan treatment resulted in a significant change in 15 of 113 proteins that are characteristic for diabetic renal damage. Similar data have been obtained in patients with vasculitis (Haubitz et al, manuscript in preparation) (85), where therapy improved/changed the vasculitisspecific protein pattern toward a normal urinary proteome.…”

“…In this study, we focused on the urinary proteome, because urine can be easily sampled and contains an array of low-molecular-weight proteins and peptides that can be analysed without additional manipulation, such as proteolysis. 3 Capillary electrophoresis coupled to mass spectrometry (CE-MS) 4,5 is now an established method, which allowed identifying peptide markers specific for a variety of diseases, such as diabetes and diabetic nephropathy, 6 graft rejection, 7 and coronary artery disease. 8 One of the core features of cardiac remodelling in hypertensive patients at risk of developing HF and patients with overt HF is related to increased interstitial deposition and cross-linking of type-I collagen.…”

“…Many of the high-throughput methods directly detect native peptides and proteins present in tissues or body fluids [20,21]. Because the termini of these naturally occurring polypeptides have not been generated by defined enzymatic cleavage and they frequently harbor PTM, direct identification of the polypeptide is challenging and often without success (e.g.…”
