High Prevalence of Scoliosis in a Large Cohort of Patients with Prader-Willi Syndrome

Abstract: The characteristics of scoliosis were investigated in a large cohort of children and adults with Prader–Willi syndrome (PWS), analysing the role of age, gender, puberty, body mass index (BMI), genotype and growth hormone therapy (GHT) on its onset and severity. A retrospective cross-sectional study was performed in 180 patients with genetically confirmed PWS (96 females), aged 17.6 ± 12 years. Eighty-five subjects (47%) were obese. One hundred and fifty subjects (83.3%) were on GHT, while 30 patients had never… Show more

“…Scoliosis is likely related to hypotonia of (paravertebral) muscles, low physical activity, and obesity ( 35 , 38 , 41 ). Additionally, late menarche and low estrogen levels might be linked to scoliosis in females with PWS ( 42–45 ), although results are inconclusive ( 40 ).…”

“…Hip dysplasia (HD) is a signi cant musculoskeletal concern in Prader-Willi Syndrome (PWS) [6,10,11] , yet the literature addressing its prevalence, diagnostic approaches, and particularly its surgical management remains scarce. Reported prevalence rates for HD in PWS vary widely, ranging from 8-30% [1,2,6,7,12,13] .…”

“…Scoliosis is present in 30%-78% of patients. 6,7 Scoliosis may affect the motor function of respiratory muscles and lead to respiratory damage. 8 Cor pulmonale and respiratory failure might be the most common cause of death and long-term complications in PWS.…”

“…8 Cor pulmonale and respiratory failure might be the most common cause of death and long-term complications in PWS. 7,9,10 Recombinant human growth hormone (rhGH) is one of the drugs used in PWS, but its effect on bone deformity is still controversial. rhGH therapy has been considered a possible risk factor for the onset of scoliosis in some studies.…”
