High Plasma Osteopontin Level and Its Relationship with Interleukin-12–mediated Type 1 T Helper Cell Response in Tuberculosis

Abstract: Osteopontin (OPN, also known as Eta-1), a noncollagenous matrix protein produced by macrophages and T lymphocytes, is expressed in granulomatous lesions caused by Mycobacterium tuberculosis infection. In the present study, we compared plasma concentrations of OPN in patients with active pulmonary tuberculosis with those of healthy control subjects and patients with sarcoidosis, another disease associated with granuloma formation. Plasma OPN levels were significantly higher in patients with tuberculosis (n = 48… Show more

“…Elevated values of circulating osteopontin have been described in a wide span of neoplastic localisations from lung to breast, colon and ovarian cancers to some non tumoural pathologies as for example tuberculosis [99]. Assaying this molecule may raise some practical concerns since we have at least three kits available on the market (from Immunobiological Laboratories, Assays Design and R&D Systems) which may not give similar results [100] but data from mesothelioma series shows similar diagnostic capabilities whichever kit is used.…”

“…Authors' Affiliations: 1 in various nonpleural malignant diseases (8 -10) and elevated serum osteopontin levels have been reported in ovarian, colon, breast, prostate, and lung cancers (11,12) or in tuberculosis (13). Osteopontin is an extracellular cell adhesion protein involved in nonmineral bone matrix formation, but is also a key cytokine in mediating type I immune responses (14), and was implicated in regulation of metastatic spread of tumor cells (15).…”

“…We assume that the quinolone treatment may be one of the factors of persistent OPN elevation. The increased plasma OPN in TB patients contributed to the disease pathology by activating the IL-12 mediated Th1 immunity (Koguchi et al 2003). It was also found that OPN expression correlates with an effective inflammatory response and contributes to human resistance against MTB (Nau et al 2000).…”
