High‐performance liquid chromatography coupled with mass spectrometry for the quantitative analysis of vinca‐alkaloids in biological matrices: a concise survey from the literature

Abstract: The bioanalysis of vinca-alkaloids has been investigated extensively. High-performance liquid chromatography coupled to ultraviolet, fluorescence or electrochemical detection have been described. During recent years liquid chromatography coupled with mass spectrometry (LC-MS) has become the first choice for the quantitative bioanalysis of the vinca anticancer agents. This paper reviews recent methods for the bio-analysis of vinca-alkaloids using LC-MS, supplemented with our own experience. We will focus on sam… Show more

“…The test showed a positive result with a half maximal inhibitory concentration (IC 50 ) value of > 32 and 350 μg ml −1 for the vinblastine that has been purified from both crude fungal extract and a crude leaf extract respectively (Figs and ). This happened when the vinca alkaloid was binding to beta‐tubulin and disruption of microtubule function during mitosis, which leads to mitosis arrest and cell death (Damen et al ., ). Jordan and Wilson () in their reports also mentioned that the anticancer activity of these alkaloids was attributed to their ability to disrupt microtubules metaphase arrest in dividing cell.…”

“…1) is a vinca alkaloid that processes strong anticancer activity and is now clinically used as an agent against breast cancer and non-small-cell lung cancer (Bonneterre et al, 2007;Delord et al, 2007). Vinorelbine is an important anticancer agent in these regions, and pharmacokinetic and pharmacological studies of it have attracted the attention of researchers (Cui et al, 2008;Damen et al, 2010;Lush et al, 2005;Tokudome et al, 2008).…”

“…One is the adsorption onto the surface of materials (Damen et al, 2010;Van Tellingen et al, 1992b), which leads to possible sample loss and to bias in the measurement. Another issue is the selection of an internal standard (IS) in mass spectrometric detection.…”

“…Traditionally, clinical and/or toxicological analyses are performed using plasma obtained by venipuncture (ter Heine et al , ; Damen et al , ). However, this sampling form requires a hospital infrastructure and is hazardous for the patient population (Boy et al , ).…”
