High Performance Liquid Chromatographic Separation and Spectral Characterization of the Pigments in Turmeric and Annatto

Abstract: High performance liquid chromatographic separations for the determination of the pigments in the food colorants, annatto and turmeric, were developed. Chromatographic analysis time for the isocratic system was 10 min and 22 min for the gradient system. The isocratic separation employed a 25 cm x 4.6 mm i.d. Zorbax ODS column with a 58/42 (v/v) mixture of waterltetrahydrofuran, THF, at 1 mL/ min. A gradient water/THF system using the same components was also developed to improve the resolution between the curcu… Show more

“…Reversed-phase chromatographic separation of the pigment mixture was achieved on a Waters pBondapak C18,lO pm column (8 mm x 100 mm) at room temperature similar to the procedure described by Rouseff (1988). Isocratic elu-tion at a flow rate of 1.5 mL/min with a mobile phase of THFIwater (40/60) and detection at 420 nm provided retention times of 15.4, 18.1 and 21.4 min for curcumin, demethoxycurcumin and bis-demethoxycurcumin, respectively.…”

“…Among them methanol does not provide the required resolution/selectivity for the separation of curcuminoids 10 . The use of tetrahydrofuran instead of acetonitrile reverses the order elution of the curcuminoids 5,8,17 . Moreover, acetonitrile is the solvent of choice because of its low wavelength transparency, polarity, and intermediate position between methanol and tetrahydrofuran 18 .…”
