High-Performance Liquid Chromatographic (HPLC) Separation and Quantitation of Endogenous Glucocorticoids after Solid-Phase Extraction from Plasma

Abstract: This study describes a method for the extraction and simultaneous measurement of cortisone, cortisol and corticosterone using dexamethasone as an internal standard. Solid-phase extraction of plasma steroids with C18 columns allows the samples to be extracted, washed and concentrated in a single step with minimal sample handling and without the use of large volumes of organic solvents. HPLC separation of the steroids is accomplished within 10 min and the individual steroid peaks are quantitated by UV detection … Show more

“…One hundred l of the supernatant were stored at Ϫ20ЊC for later corticosterone (CORT) determination and 100 l at Ϫ80ЊC for catecholamine [noradrenaline (NA) and adrenaline (A)] measurements. For plasma CORT assessments, a reversedphase, high-performance liquid chromatography (HPLC) technique was used as described in Dawson, Kontur, & Monjan (1984). NA and A were extracted from the samples by using an extraction system as described in Smedes, Kraak, & Poppe (1982).…”

“…Plasma levels of insulin, leptin and ACTH were measured by commercial radioimmunoassay kits (Linco Research; RI-13K, RL-83K and ACTH RIA kit, Nichols Institute Diagnostics, San Juan Capistrano, CA, USA). Plasma levels of corticosterone were measured using HPLC with UV detection according to Dawson et al, [22].…”
