A micellar electrokinetic capillary chromatographic (MEKC) procedure has been developed for the separation and determination of dioxouranium (VI), iron(III), copper(II), and nickel(II) using bis(salicylaldehyde)propylenediimine (H2SA2Pn) as chelating reagent with a total run time of <3 min. Sodium dodecyl sulphate (SDS) was used as micellar medium at pH 8.1 with sodium tetraborate buffer (0.1 M). Uncoated fused silica capillary with effective length 38.8 cmx75 microm id was used with an applied voltage of 30 kV and photo-diode array detection at 228 nm. Linear calibrations were established within 0.045-1000 microg/mL of each element with detection limit within 15-122 ng/mL. The method was applied to the analysis of spring water and rock samples. The presence of uranium in rock and spring water samples was established within 1.58-1739.3 microg/g and 0.047-0.712 microg/mL with relative standard deviation within 0.9-2.1% and 1.3-2.6% respectively. Uranium ore and water samples were also assayed by the standard addition technique. Recovery of uranium was >98% with RSD up to 2.7%. Copper, nickel, and iron in their combined matrix were concurrently determined within RSD 0.6-3.6% (n=5) and the results obtained were compared with those of flame AAS.

A capillary liquid chromatography with UV detection (CLC-UV) system has been developed for determining platinum-based antitumor drugs (e.g., cisplatin, carboplatin, and nedaplatin) in plasma based on the pre-column derivatization of platinum with N,N-diethyl dithiocarbamate (DDTC). The chelated platinum separation was carried out on a capillary column (Inertsil ODS-3, 150 mm 0.3 mm i.d., 3 µm) using an acetonitrile-water mixture (8 : 2, v/v) as a mobile phase that flowed at 5.0 µL/min. Detection was carried out by absorbance at 254 nm. Chromatographic peak height was found to be linearly related to the spiked concentration of nedaplatin in the blank control plasma from 5.0 ng/mL to 15 µg/mL (r 2 >0.998). The repeatability (n 5) of the chromatographic peak height for 2.5 µg/mL nedaplatin was 2.6% relative standard deviation (R.S.D.). The CLC-UV system, which required only 20 µL of plasma sample, was applied to the determination of total and free form platinum-based antitumor drugs in plasma after injection into rats. The recovery rates (n 5) of total and free form nedaplatin in plasma were 98% and 99%, respectively, and these repeatability were 2.4% R.S.D. and 3.1% R.S.D., respectively. In addition, the recovery rates (n 5) of total and free form carboplatin in plasma were 99% and 99%, respectively, and these repeatability were 2.9% R.S.D. and 0.24% R.S.D., respectively. The concentration-time profiles of total and free form nedaplatin in rat plasma were monitored to determine the pharmacokinetic parameters.Key words platinum-based antitumor drug; plasma; capillary column; liquid chromatography Platinum-based antitumor drugs, including cisplatin, carboplatin, oxaliplatin, and nedaplatin are used to treat various types of cancers, such as sarcomas, carcinomas, etc. Nedaplatin, cis-diammineglycolatioplatinum, is a second-generation platinum agent that is approximately 10 times as soluble in water as cisplatin. [1][2][3][4] As such, nedaplatin is considered to have more pronounced activity against solid tumors, but less nephrotoxicity and gastrointestinal toxicity than cisplatin.

Sample preparation is a major task in a regulated bioanalytical laboratory. The sample preparation procedure significantly impacts assay throughput, data quality, analysis cost, and employee satisfaction. Therefore, selecting and optimizing an appropriate sample preparation method is essential for successful method development. Because of our recent expertise, this article is focused on sample preparation for high-performance liquid chromatography with mass spectrometric detection. Liquid chromatography with mass spectrometric detection (LC-MS) is the most common detection technique for small molecules used in regulated bioanalytical laboratories. The sample preparation technologies discussed are pre-extraction and post-extraction sample processing, protein precipitation (PPT), liquid-liquid extraction (LLE), offline solid-phase extraction (SPE), and online solid-phase extraction. Since all these techniques were in use for more than two decades, numerous applications and variations exist for each technique. We will not attempt to categorize each variation. Rather, the development history, a brief theoretical background, and selected references are presented. The strengths and the limitations of each method are discussed, including the throughput improvement potential.
