High‐level expression of the ER‐MP58 antigen on mouse bone marrow hematopoietic progenitor cells marks commitment to the myeloid lineage

Abstract: Studies on the early events in the differentiation of the nonspecific immune system require the identification and isolation of myeloid-committed progenitor cells. Using the monoclonal antibodies (mAb) ER-MP12 and ER-MP20, generated against immortalized macrophage precursors, we have shown previously that the earliest macrophage colony-stimulating factor (M-CSF)-responsive cells in the bone marrow have the ER-MP12hi 20- phenotype. In addition, we found that the ER-MP12hi 20- subset (comprising about 2 % of tot… Show more

“…Indeed, the local pancreas DC precursor has a similar phenotype as blood monocytes, except for the lower CD11c expression on the Ly6C low cells and is expressing ER-MP58, which is a marker for both myeloid precursors in the BM and peripheral blood monocytes [15]. Upon GM-CSF stimulation the local ER-MP58 1 cells isolated from fetal pancreas displayed a high proliferative activity.…”

“…However, some non-myeloid cells can express this marker at low levels [15]. Isolated ER-MP58 1 cells from the pancreas were used in cultures with GM-CSF and developed into DCs.…”

“…We also measured the expression of the hematopoietic markers ER-MP12, ER-MP20, and ER-MP58, which recognize hematopoietic precursors and have previously been used as markers of monocytic differentiation. “…However, heterogeneity of monocyte populations has been poorly understood, particularly in comparison with lymphoid and dendritic cell populations. Several monocytic/myeloid precursor populations in BM have been defined by cytochemical methods (2)(3)(4)(5) as well as antigen expression (5)(6)(7)(8)(9)(10), and recent studies have begun to define subpopulations of monocytes in murine peripheral blood (PB) (11)(12)(13). These populations have been defined by differential expression of several markers, such as CX 3 (12,13).…”
