High Interferon Signature Leads to Increased STAT1/3/5 Phosphorylation in PBMCs From SLE Patients by Single Cell Mass Cytometry

Abstract: The establishment of an “interferon (IFN) signature” to subset SLE patients on disease severity has led to therapeutics targeting IFNα. Here, we investigate IFN signaling in SLE using multiplexed protein arrays and single cell cytometry by time of flight (CyTOF). First, the IFN signature for SLE patients (n=81) from the Stanford Lupus Registry is determined using fluidigm qPCR measuring 44 previously determined IFN-inducible transcripts. IFN-high (IFN-H) patients have increased SLE criteria and renal/CNS/immun… Show more

“…Furthermore, in contrast to humoral responses, impaired T cell induction was also observed in patients with SLE not taking immunosuppressive medication. These effects may be due to intrinsic SLE-related impairments in T cell function (32,33), highlighting the need for further studies aimed at enhancing T cell responses and optimizing vaccine efficacy in those with SLE. Evaluation of SLE flares following mRNA vaccination is particularly important, given the reported vaccine hesitancy among persons with this condition.…”

“…The authors considered stimulating whole PBMC samples separately for the T-cell/NK cell or the B-cell/monocyte panels. Although, we found multiple examples for activation of whole PBMC samples before performing mass cytometry (90)(91)(92)(93). Even in the scope of our research, autoimmunity field, this method was applied.…”
