High Frequency of Thyroid Tumor Induction by N‐Methyl‐N′‐nitro‐N‐nitrosoguanidine in the Hermaphroditic Fish Rivulus marmoratus

Abstract: In the self‐fertilizing hermaphroditic fish, Rivulus marmoratus, the susceptibility to tumor induction by N‐methyl‐N‐nitro‐N‐nitrosoguanidine(MNNG) was evaluated. Seven‐day‐old fish larvaewere exposed for 2 h toMNNG at concentrations ranging from 5 to 25 ppm in a static water bath. The exposed fish were observed at 2 and 4 monthsafter carcinogen treatmentto assess tumor development. Within 4‐ months after25 ppmMNNG exposure, nearly all fish developed thyroid tumors. The tumor incidenceswere dose‐ and time‐depe… Show more

“…With scale removal, the scale-associated cells are also removed, and there is proliferation of the scale pocket lining cells into layers with the production of scale matrix. There is also a close association between the scale-forming cells and the epidermis, which guarantees that only 1 scale will reform within the scale pocket (2,39,44 (40) and thyroid hyperplasia and ectopic follicles in liver from DEN (21) were reported in rivulus. In the present study, all of the MNNG-induced thyroid neoplasms were seen in males.…”

“…Thyroid neoplasms were seen in rivulus exposed at 1 wk of age, but neoplasms in other tissues were not induced (145). Osteogenic neoplasms were reported in coho salmon (Oncorhynchus kisutch) exposed as embryos (21 ) and in channel catfish (41).…”

“…One case of follicular-derived thyroid cancer was observed with DMBA treatment, but none were observed with MNNG treatment. Interestingly, thyroid cancer has been reported at higher levels in other fish species treated with MNNG (Park et al 1993). These and other chemical carcinogenesis studies demonstrate that chemical mutagens retain the ability to cause malignant transformation in zebrafish tissues.…”
