High Frequency of Mutations in thePIK3CAGene Helical and Kinase Coding Regions in a Group of Iranian Patients with High-Grade Glioblastomas: Five Novel Mutations

Abstract: Glioblastoma multiform (GBM; World Health Organization (WHO) grade IV) and anaplastic astrocytomas (AA; WHO grade III) are highly aggressive and lethal astrocytic brain tumors. To detect cancer-specific somatic mutations in two hot-spot regions of PIK3CA gene, the helical and kinase domains (encoded by exons 9 and 20, respectively) in GBM and AA, the authors examined the respective sequences 31 paraffin-embedded samples (23 GBM and 8 AA). The samples were obtained from a group of Iranian patients affected with… Show more

“…Eighty six up regulated proteins were enriched in progesterone-mediated oocyte maturation, which is an important process in cell cycle regulation, as its dysregulation can lead to uncontrolled proliferation of cancer cells 65 . In addition, studies have shown that progesterone-mediated oocyte maturation is related to the pathogenesis of glioma 66 .…”

“…Annibali et al, (2014) found that Myc inhibition reduces proliferation, increases apoptosis, and, remarkably, elicits the formation of multinucleated cells that then arrest or die by mitotic catastrophe, revealing a new role for Myc in the proficient division of glioma cells. Some results confirmed that PIK3CA mutations occurred in a significant number of human glioblastomas, making it a promising target for therapy, particularly for primary glioblastomas (Weber et al, 2011;Derakhshandeh-Peykar et al, 2012). A meta-analysis by Xiao et al (2012) provided direct and strong evidences that mutations in the PTEN gene were correlated with the poor prognosis of glioma patients (Han et al, 2014).…”

“…Mutation p.M1043I was described in previous studies as oncogenic [9]. Mutation p.N1044D has been detected only in a few cases of large intestine carcinoma, endometrial carcinoma and glioblastoma so far [34][35][36]. Using in silico tool PROVEAN, we evaluated its effect on the protein function as neutral (PROVEAN score -0.458).…”
