High Density Lipoproteins and Arteriosclerosis

Abstract: Abstract-High density lipoprotein (HDL) cholesterol is an important risk factor for coronary heart disease, and HDL exerts various potentially antiatherogenic properties, including the mediation of reverse transport of cholesterol from cells of the arterial wall to the liver and steroidogenic organs. Enhancement of cholesterol efflux and of reverse cholesterol transport (RCT) is considered an important target for antiatherosclerotic drug therapy. Levels and composition of HDL subclasses in plasma are regulated… Show more

“…It is well accepted that the removal of cholesterol from peripheral cells by HDL involves signal transduction systems in which HDL receptors at the cell surface are believed to transmit the signal to intracellular effectors (22,23). In this report, we demonstrate that the treatment of stimulated normal fibroblasts with apoA-I for short periods of time (Յ45 min) induces ABCA1 phosphorylation activity in a concentration-dependent manner (Fig.…”

“…It is postulated that this is due to the role of HDL in reverse cholesterol transport from the cells of the arterial wall (1,2). Reverse cholesterol transport involves the HDL-mediated transport of cholesterol from extrahepatic tissues to the liver, allowing for cells to rid themselves of excess cholesterol and maintain cholesterol balance.…”

“…4B), the most important acceptors of cell-derived phospholipids and cholesterol (reviewed in Ref. 48). Furthermore, another study has shown that proteolysis leads to dissociation of apoA-I from HDL, especially on the less buoyant HDL 3 , and produces an unstable HDL population (49).…”
