High concentrations of atrial natriuretic peptide and brain natriuretic peptide in rat pericardial fluid and their reduction by reserpine in vivo

Abstract: We determined concentrations and molecular sizes of natriuretic peptides in rat pericardial fluid and plasma by use of specific radioimmunoassays (RIA) and gel filtration HPLC. Our study shows that pericardial fluid forms a local extracellular storage of immunoreactive (ir) atrial natriuretic peptide (irANP) and brain natriuretic peptide (irBNP) near the heart where these peptides can be found in high concentrations in vivo. The concentrations of irANP, irBNP and NH2-terminal fragment of proANP (irNT-proANP) i… Show more

“…This is observed, for instance, for endothelin-1 (Horkay et al, 1995), FGF-2 (Corda et al, 1997), and atrial natriuretic peptide (Amano et al, 1993;Corda et al, 1997) in heart surgery patients. Also, in rats, concentrations of atrial natriuretic peptide in pericardial fluid are higher than in plasma (Klemola et al, 1995). The present study shows that these high pericardial fluid/plasma concentration ratios not only are a matter of local syntheses of the agents in the heart or in the mesothelial cells of the pericardium (Eid et al, 1994;Mebazaa et al, 1998), but can also be attributed to the relatively slow clearances of these agents, once "trapped" into pericardial space.…”

“…The existence of a high number of vesicles of micropinocytosis suggests an active exchange of solutes with the pericardial cavity. This may be important since the pericardial fluid contains atrial natriuretic peptides (ANP) in mammals (Klemola et al, 1995), and ANP‐binding sites have recently been described in the bulbus arteriosus of C. hamatus (Cerra et al, 1997). Pinocytotic activity of the visceral pericardium may control the passage of these and other substances, thus modulating a number of heart activities.…”
