High cell density cultivation and recombinant protein production with Escherichia coli in a rocking-motion-type bioreactor

Abstract: BackgroundSingle-use rocking-motion-type bag bioreactors provide advantages compared to standard stirred tank bioreactors by decreased contamination risks, reduction of cleaning and sterilization time, lower investment costs, and simple and cheaper validation. Currently, they are widely used for cell cultures although their use for small and medium scale production of recombinant proteins with microbial hosts might be very attractive. However, the utilization of rocking- or wave-induced motion-type bioreactors… Show more

“…3). This is a common trait of most bacteria during hydrophobic wastewater treatment (Frolund et al, 1996), which is involved in the regulation of organic pollutants' transport across the bacterial cell membrane and a very complex metabolic pathway (Glazyrina et al, 2010). Additionally, it opens up the possibility of using both bacteria to produce proteins with distinctive properties simply by manipulation of the cultivation conditions.…”

“…After 16 hours of expression, we obtained a biomass yield of 40 g WCW/1 L culture. Our procedure for highyield protein expression obtained in high-density bacterial cultures has resulted in similar protein production rates reported in other studies concerning expression of alcohol-dehydrogenase [16], even labeled eukaryotic proteins [17] or for expression of genes from heterologous biosynthetic pathways [18].…”

“…Glazyrina et al demonstrated the feasibility of using the BIOSTAT CultiBag RM system for the cultivation of recombinant Escherichia coli. A dry cell weight of 10 g L -1 was obtained when using an internal feeding system (EnBaseÒ, BioSilta Oy, Oulu, Finland) [8]. A twofold increase in the cell density was achieved by employing a fed-batch procedure with additional oxygen sparging.…”
