High animal fat intake enhances prostate cancer progression and reduces glutathione peroxidase 3 expression in early stages of TRAMP mice

Abstract: Dietary fat enhances prostate cancer progression, possibly by suppressing GPx3 expression and increasing proliferation of prostate intraepithelial neoplasia (PIN) epithelial cells.

“…Thus, the proven anti-inflammatory properties of specific serenoa, pomegranate and pumpkin extracts [37][38][39]42] which are not directly shared by finasteride, come to be all the more relevant. This has been confirmed last month also in an obese mouse model [43] further paving the way to a wider systemic metabolic understanding of the disease [44][45][46][47]. In favour of such integrated phyto-pharmaceutic approach, it is worthwhile reporting that components such as specific pumking seed, red clover isoflavones, serenoa and pomegranate extracts have been shown to have antiandrogenic activity by dosedependently blocking the binding of DHT besides am anti-aromatase and anti-5-α-reductase Type II action [38,[48][49][50][51].…”

“…It is well known that glutathione peroxidases are among the most important reactive oxygen species scavengers that protect cells from oxidative damage (36). Downregulation of GPx3 by hypermethylation has been revealed in numerous types of cancer (37)(38)(39)(40)(41)(42)(43)(44)(45), including prostate cancer (11,46,47). In prostate cancer, a negative correlation between GPx3 expression levels and poor clinical outcomes has been demonstrated (47,48).…”

“…In prostate cancer, a negative correlation between GPx3 expression levels and poor clinical outcomes has been demonstrated (47,48). Forced expression of GPx3 in prostate cancer cell lines is able to suppress colony formation and cellular proliferation (11,47,49). Xenografts with higher GPx3 expression levels have resulted in reduced tumor volume, metastasis and animal mortality (47).…”

“…Down-regulation of GPx3 increases these cell's vulnerability to oxidative damages. Conditions predisposed to prostate cancer, such as high animal fat diet in TRAM mice and loss of tumor suppressor Nkx3.1, has been found accompanied with decrease in GPx3 expression in animal (Chang et al 2014), (Ouyang et al 2005).…”
