HIFs, angiogenesis, and cancer

Abstract: Tumor hypoxia was first described in the 1950s by radiation oncologists as a frequent cause of failure to radiotherapy in solid tumors. Today, it is evident that tumor hypoxia is a common feature of many cancers and the master regulator of hypoxia, hypoxia-inducible factor-1 (HIF-1), regulates multiple aspects of tumorigenesis, including angiogenesis, proliferation, metabolism, metastasis, differentiation, and response to radiation therapy. Although the tumor hypoxia response mechanism leads to a multitude of … Show more

“…Angiogenin appears to be a crucial stimulant for the angiogenic process to allow tumor growth beyond a few millimetres as well as the development of metastasis [20]. This was confirmed in our study, angiogenin was highly expressed in invasive stage (II-III) (100%) than superficial stage (0-I) (43.33%) at P <0.05.…”

“…Importantly, IL-37 expression and secretion is induced by hypoxia, a well-known proangiogenic conditions that stimulates angiogenesis and vascular alterations. 25 Knockdown of HIF-1α expression inhibited hypoxia-induced upregulation of Il37 mRNA and IL-37 protein levels, indicating that IL-37 expression and secretion are regulated by proangiogenic hypoxic signaling. Interestingly, the principle angiogenic factor VEGF did not have any effect on the expression of IL-37.…”

“…Stabilization of hypoxia-inducible factor-alpha (HIF-α) transcription factors by low oxygen tension in developing tumors is arguably the most important determinant of solid tumor neovascularization (36). MIF was first linked to tumor hypoxic responses when it was reported that MIF mRNA levels are induced by low oxygen tensions in human squamous carcinoma cell lines (37).…”
