HIF‐1α Contributes to Hypoxia‐induced VSMC Proliferation and Migration by Regulating Autophagy in Type A Aortic Dissection

Ben Huang,
Nan Chen,
Zhenhang Chen
et al.

Abstract: Type A aortic dissection (AD) is a catastrophic cardiovascular disease. Hypoxia‐inducible factor‐1α (HIF‐1α) and autophagy are reported to be upregulated in the AD specimens. However, the interaction between HIF‐1α and autophagy in the pathogenesis of AD remains to be explored. HIF‐1α and LC3 levels are evaluated in 10 AD and 10 normal aortic specimens. MDC staining, autophagic vacuoles, and autophagic flux are detected in human aortic smooth muscle cells (HASMCs) under hypoxia treatment. CCK‐8, transwell, and… Show more

“…Furthermore, we showed that chronic IH-induced autophagy and mitophagy, is not observed in HIF-1 α heterozygous mice demonstrating that IH-induced mitochondrial remodeling depends on HIF-1 upon IH. This aligns with the role of HIF-1 in inducing autophagy by activating the transcription of genes, including BNIP3 27,28 , one of the main autophagic receptor activated by hypoxia 30 . HIF-1 has also been shown to positively regulate parkin-mediated mitophagy in acute kidney injury 31 and in diabetes 32 .…”

“…Because chronic IH alters mitochondrial function and leads to cardiomyocytes apoptosis [13][14][15] , we first investigated if IH induced mitochondrial remodeling by evaluating autophagy and mitophagy. Since HIF-1 is recognized to activate genes encoding for the autophagic process 27,28 , we also questioned if IH-induced mitochondrial remodeling depends on HIF-1.…”
