Hierarchically Structured Polystyrene-Based Surfaces Amplifying Fluorescence Signals: Cytocompatibility with Human Induced Pluripotent Stem Cell

Abstract: An innovative multi-step phase separation process was used to prepare tissue culture for the polystyrene-based, hierarchically structured substrates, which mimicked in vivo microenvironment and architecture. Macro- (pore area from 3000 to 18,000 µm2; roughness (Ra) 7.2 ± 0.1 µm) and meso- (pore area from 50 to 300 µm2; Ra 1.1 ± 0.1 µm) structured substrates covered with micro-pores (area around 3 µm2) were prepared and characterised. Both types of substrate were suitable for human-induced pluripotent stem cell… Show more

“…Phase Inversion Modification of the HU Surface. Similar to the case of PS, 40 the addition of HU to the modification solution allowed for preparation of new surface texture types. From a physicochemical point of view, this approach corresponds to the phase inversion described in the literature.…”

“…We found a strong correlation between these two, in the sense that the morphology of the cells is severely influenced by the environmental conditions, which act as a key factor in cellular differentiation [ 54 , 56 ]. The architecture of the cell culture substrate has been previously shown to influence cells differentiation [ 57 , 58 , 59 ]. Here, the elongated and flattened polygonal shape of the OLN 93 cells grown on the glass substrate ( Figure 1 a), is associated with the differentiation of cells towards the mature form in which they fulfill their physiological function of producing the myelin sheath [ 56 ].…”
