HHLA2 and PD-L1 co-expression predicts poor prognosis in patients with clear cell renal cell carcinoma

Abstract: BackgroundAlthough clear cell renal cell carcinoma (ccRCC) is well known as a highly immunogenic tumor, only a small subset of patients could benefit from current immunotherapy, which might be due to the heterogeneity of immune microenvironment in ccRCC. So, it is meaningful to explore novel immunotherapy or combination therapy for improving therapeutic efficacy. HHLA2, a newly discovered B7 family member, is prevalently expressed in numerous tumors, including ccRCC. This study aimed to investigate the prognos… Show more

“…According to Teng's theory, it may be much more rational to classify by inflammatory cells via the combination PD-L1 and CMTM6 or in combination with other CMTM family members in HCC [34]. Consistent with previous study, our results demonstrated that in inflammatory cell-positive groups, CMTM6/ PD-L1 coexpression displayed significant prognostic value for DFS and OS in MTM subtype [35]. These results suggested that both CMTM6 and PD-L1 perform immunosuppressive functions that partly depend on suppression of the inflammatory cell function.…”

“…CD8+ or Foxp3+ TILs were calculated in the same manner as described in the previous study. 16 In brief, the average of five independent high-power fields (400×), which represented the densest lymphocytic infiltrates, was used to reflect the extent of T cell infiltration.…”

“…Human endogenous retrovirus-H long terminal repeat-associating protein 2 (HHLA2), also known as B7H7 or B7-H5, is a newly discovered member of the B7 family. [ 13 ] HHLA2 effectively suppresses the activity of CD4+ and CD8+ T cells and reduces the expression of T cell cytokines, such as interferon-γ, tumor necrosis factor-α, interleukin (IL)-5, IL-10, IL-13, IL-17α and IL-22. [ 14 ] In addition, HHLA2 is an inducer of the inflammatory response of antigen-presenting cells.…”
