Heterologous Expression of Bovine Rhodopsin inDrosophilaPhotoreceptor Cells

Abstract: Drosophila photoreceptors express a vertebrate rhodopsin as a functional visual pigment, but the expression does not activate the Drosophila phototransduction pathway. The system allows the characterization and comparison of vertebrate and invertebrate visual pigment properties in a common cell type.

“…Altogether, these experiments demonstrate that the Rh1 function in promoting rhabdomere and RTW morphogenesis is highly conserved. Because bovine rhodopsin cannot activate the heterotrimeric G proteins (Gqe) involved in invertebrate phototransduction (Ahmad et al, 2006), this also confirms that the Rh1 function in rhabdomere and RTW morphogenesis is independent of heterotrimeric G-protein function.…”

“…3C); however, this base was distorted and small, indicating that human melanopsin does not fully support rhabdomere development. Finally, we expressed the bovine ciliary opsin (rhodopsin) in ninaE l17 eyes (Ahmad et al, 2006). It is known that bovine rhodopsin can also rescue the early developmental failure in rhabdomere morphogenesis (Ahmad et al, 2007), and the corresponding photoreceptors indeed appeared identical to those in wild-type flies 1566 Journal of Cell Science 124 (9) (Fig.…”

“…For this analysis, we created Drosophila strains in which a transgene was the only rhodopsin expressed in the R1-6 cells and there was no light response from R7 and R8 photoreceptors (Ahmad et al, 2006). In the absence of a rhodopsin transgene, no light response is evoked using 600-350 nm light stimuli, with intensities adjusted so that similar photon contents were delivered.…”

“…Transgenic D. melanogaster expressing A. aegypti Aaop8 and Aaop10 in R1-6 photoreceptors The Aaop8 ORF was directionally cloned into a modified pCaSpeR4 transformation vector, in which the polylinker region was replaced with the Drosophila Rh1 rhodopsin (ninaE) promoter, and a 0.7 kb 3′ untranslated region of the ninaE gene (Ahmad et al, 2006). For Aaop10, we modified the pCaSpeR4 vector by adding an attR1-ccdB-attR2 Gateway recombination site, then the Aaop10 ORF plasmid described above was inserted into this plasmid using Gateway technology (Life Technologies, Grand Island, NY, USA).…”

“…Therefore only the activity of Aaop7 or the Rh1 control rhodopsin was represented in the ERG response (Ahmad et al, 2006). The Aaop7-and the Rh1-expressing Drosophila were then subjected to ERG analysis as described previously (Hu et al, 2014).…”
