Heterologous downregulation of vasopressin type 2 receptor is induced by transferrin

Abstract: Vasopressin (VP) binds to the vasopressin type 2 receptor (V2R) to trigger physiological effects including body fluid homeostasis and blood pressure regulation. Signaling is terminated by receptor downregulation involving clathrin-mediated endocytosis and V2R degradation. We report here that both native and epitope-tagged V2R are internalized from the plasma membrane of LLC-PK1 kidney epithelial cells in the presence of another ligand, transferrin (Tf). The presence of iron-saturated Tf (holo-Tf; 4 h) reduced … Show more

“…20,21 A similar pattern was observed with AG-490, a combined JAK-2 and EGFR inhibitor. The role of the JAK/STAT signaling pathway was ruled out, because TG-101348, a selective JAK-2 inhibitor, did not induce AQP2 membrane accumulation.…”

“…We then used our GFP-tagged V2R cell model. At baseline, V2R accumulates in the plasma membrane, and, with VP treatment, we observed a marked internalization of V2R-GFP, as previously described (50,67). This suggests that the clathrin-mediated endocytosis process that is necessary for receptor downregulation after ligand binding still occurs when Arp2/3 activity is inhibited (Fig.…”

“…We then tested whether overexpression of XLαs affects internalization of the type 2 vasopressin receptor (V2R), another GPCR that internalizes via clathrin-mediated endocytosis (45,46). HEK293T cells were cotransfected with XLαs-GFP and V2R cDNA and treated with tetramethylrhodamine-labeled vasopressin (VP-TMR) for 5 min.…”
