Heterogenous graft rejection pathways in class I major histocompatibility complex-disparate combinations and their differential susceptibility to immunomodulation induced by intravenous presensitization with relevant alloantigens.

Abstract: SummaryThe present study investigates the heterogeneity of graft rejection pathways in class I major histocompatibility complex (MHC)-disparate combinations and the susceptibility of each pathway to immunomodulation induced by intravenous presensitization with alloantigens . Depletion of CD8+ T cells was induced by repeated administration of anti-CD8 monoclonal antibody. CD8+ T cell-depleted mice failed to generate anti-allo class I MHC cytotoxic T cell (CTL) responses but exhibited anti-allo class I MHC T cel… Show more

“…One day before each immunization, the mice were intraperitoneally given 200 g/mouse of anti-CD4 (GK1.5) or anti-CD8 (2.43) monoclonal antibody in PBS (total, three times/ mouse). The depletion of CD4 ϩ and CD8 ϩ T cells in mice 7 days after the last injection of respective monoclonal antibodies was Ͼ98% when evaluated by flow cytometric analysis of their splenocytes as described previously (15).…”

“…C D4 T helper cells play a pivotal role in orchestrating allograft rejection and provide help for the generation of both CD8 T effector cells and alloantibody production by B cells (1)(2)(3)(4)(5). Understanding the nature of the cell-cell interactions involved when CD4 T cells provide help for T cell and alloantibody mediated effector responses is important for developing more rational approaches for preventing allograft rejection.…”

“…More recently, Kitagama et al (35) have reported that treatment of the recipient with either anti-CD8 or anti-CD4 enhances donor-specific response reduction either as measured in an MLR or as assessed by skin graft survival. Anti-CD4 was effective only if administered before allogeneic cell injection.…”
