Heterogeneous nuclear ribonucleoprotein A/B: an emerging group of cancer biomarkers and therapeutic targets

Abstract: Heterogeneous nuclear ribonucleoprotein A/B (hnRNPA/B) is one of the core members of the RNA binding protein (RBP) hnRNPs family, including four main subtypes, A0, A1, A2/B1 and A3, which share the similar structure and functions. With the advance in understanding the molecular biology of hnRNPA/B, it has been gradually revealed that hnRNPA/B plays a critical role in almost the entire steps of RNA life cycle and its aberrant expression and mutation have important effects on the occurrence and progression of va… Show more

“…HnRNPAB is a member of the hnRNP family. Previous studies have shown that hnRNPAB is dysregulated in multiple tumors, which is associated with various malignant phenotypes of tumors (10)(11)(12)31). However, the expression and role of hnRNPAB in NSCLC remain unclear.…”

“…Moreover, the exact location of site cg15658376 was on chr5, expanding across a large region with distinguished H3K27Ac label, suggesting methylation modification of cg15658376 might silence a possible enhancer of gene heterogeneous nuclear ribonucleoprotein A/B (HNRNPAB). HNRNPAB is closely associated with pre‐mRNAs within the nucleus and is known to regulate pre‐mRNA processing as well as other crucial aspects of mRNA metabolism and transport 26 . hnRNPA0, a main subtype of hnRNP, was found to be a strong promoter for EC 27 .…”

“…22,23 These small molecules can be designed to target an array of actors within the context of splicing, from spliceosome components to diverse trans-acting splicing factors, and even RNA targets themselves, including both regulatory and precursor messenger RNAs. [24][25][26][27][28] In particular, the task of designing small molecules that specifically bind to RNA and modulate splicing is a substantial challenge. Indeed, RNA molecules have markedly fewer hydrophobic surfaces relative to proteins and they present more restricted binding Fig.…”
