Heterogeneous expression and functional evaluation of in silico characterized recombinant OmpC of Salmonella Typhimurium as a functional poultry vaccine to eradicate zoonotic transmission

Abstract: Salmonellosis is one of the major global health concerns leading to millions of deaths annually. The present vaccines not being up to the mark necessitate the need for the development of new generation vaccines. Outer membrane proteins (Omps) of several Gram negative bacteria have been investigated and found to be immunogenic and protective. The present study explores the potential of a major porin protein (OmpC) of Salmonella Typhimurium, as a vaccine candidate. The OmpC 3D structure and its potential to bind… Show more

“…The calcium phosphate nanoparticles along with chitosan are the new generation adjuvants which show a balanced Th1 and Th2 immune response [13–15, 33, 34]. Outer membrane proteins (Omps) and outer membrane vesicles (OMVs) have been used as candidate vaccine antigen against Salmonella serovars [2, 35, 36]. Omps of Salmonella were also tested as injectable vaccine preparation and also as oral vaccine by microencapsulation in poly lactic-co-glycolic acid (PLGA).…”

“…Omps of Salmonella were also tested as injectable vaccine preparation and also as oral vaccine by microencapsulation in poly lactic-co-glycolic acid (PLGA). The micro encapsulated Omps depicted promising immune potential through both the routes [29, 35, 37]. In this work, we studied the characteristics of calcium phosphate nanoparticles adjuvanted Omps vaccine formulation and their toxicity.…”

“…When required, the above media were supplemented with ampicillin (100 μg/mL) and kanamycin (50 μg/ mL) antibiotics [Himedia, India]. Antiserum against Omps of S. Typhimurium raised in rabbit was procured from Animal Biotechnology Centre, GBPUA&T, Pantnagar (JHA et al, 2015).…”

“…The outer membrane proteins (Omps) from Salmonella have been proven to have good immunogenicity (LEE et al, 2010;GIL-CRUZ et al, 2009). Since that discovery, Salmonella Omps have been characterized (HAMID and JAIN, 2008) and targeted for protection (JHA et al, 2015;SAXENA et al, 2012;BHAT and JAIN, 2010). Omp28 was isolated independently from S. Typhi by BHATNAGAR et al (1982) and DE ANDRADE et al (1998).…”
