Heterogeneous dimer peptide-conjugated polylysine dendrimer-Fe3O4 composite as a novel nanoscale molecular probe for early diagnosis and therapy in hepatocellular carcinoma

Abstract: A novel nanoscale molecular probe is formulated in order to reduce toxicity and side effects of antitumor drug doxorubicin (DOX) in normal tissues and to enhance the detection sensitivity during early imaging diagnosis. The mechanism involves a specific targeting of Arg-Gly-Asp peptide (RGD)-GX1 heterogeneous dimer peptide-conjugated dendrigraft poly- l -lysine (DGL)–magnetic nanoparticle (MNP) composite by α v β 3 -integrin/vasculature endot… Show more

“… 41 , 42 , 43 Tailored SPIO/USPIO nanoparticles can also specifically target tumor cells through their coupling with aptamers, peptides and antibodies. 44 , 45 , 46 , 47 Recently, a bi-specific USPIO imaging probe was developed for HCC diagnosis by simultaneously conjugating AFP and GPC3 antibodies. The system has already shown higher sensitivity and specificity than single antibody-labeled or unlabeled USPIO in vitro .…”

“…1 Chemotherapy is still a major clinical method for cancer treatments. 2 However, the traditional chemotherapeutic drugs are generally nonspecific and have no ability to discriminate between cancer cells and normal cells, which frequently causes systemic side effects.…”

“…This targeting strategy draws lessons from viruses, which engage in multivalent interactions with multiple membrane receptors before entering host cells 45 . Doxorubicin (DOX)-loaded dendrigraft poly-l-lysine (DGL)-MNP modified with RGD- and GX1-peptides, which target the vascular endothelial growth factor and integrin receptors, respectively, demonstrated significantly enhanced HepG2 tumor accumulation of these dual-ligand probes compared to the single-ligand probes 46 . In addition, groups treated with these dual-ligand probes demonstrated significantly higher tumor inhibition rate (78.5%) 18 days after treatment compared to groups treated with free DOX (39.6%), MNP-DGL-RGD-GX1 (32.7%), MNP-DGL-RGD-DOX (65.8%), and MNP-DGL-GX1-DOX (68.4%).…”
