Heterogeneity of neuropeptide Y immunoreactivity in patients with pheochromocytoma: influence on the diagnostic power of measuring plasma NPY using antisera with different specificities

Abstract: The nature of NPY-like immunoreactivity (NPY-LI) was investigated in plasma and tumour tissue of 1 7 pheochromocytoma patients by HPLC, gel filtration and isoelectric focusing using two radioimmunoassays (RIAs) directed against the C- and N-terminals of NPY respectively. The two RIAs gave similar results in pheochromocytomas: 86% of cases had higher NPY-LI concentrations than those found in normal adrenal glands and NPY-LI behaved like authentic human NPY during gel filtration and HPLC. Assessed by isoelectric… Show more

“…Elevated systemic NPY levels correlate with adverse tumor phenotype in Ewing's sarcoma and neuroblastoma 27,40 . There are associations of elevated plasma NPY with metastasis in pheochromocytoma 41,42 . An increase in NPY expression in tissues is an early event in PCa development 43 .…”

“…M-8701, Milab, Malmö, Sweden: dilution 1:640) according to the method described above. To study the possible coexistence of PACAP and NPY the elut ion-restaining technique or consecutive staining of adjacent cryo stat sections for PACAP and NPY were performed using an NPY antiserum obtained by immunizing rabbits against synthetic porcine NPY (Peninsula, Belmont, Calif., USA) conjugated with bovine serum albumin (code K02B; dilution 1:640) [15]. For control, the NPY antiserum was incubated with 10-100 pg of NPY before applying to the sections.…”

“…O NPY like-immunoreactivity (NPY-LI) sofre um incremento nos níveis circulantes durante a manipulação tumoral cirúrgica, reduzindo em mais de 70% dos casos sua concentração em cerca de uma hora após a retirada do tumor. Aproximadamente 86% dos feocromocitomas apresentam elevação dos níveis de NPY-LI tecidual em relação as adrenais normais, e, apesar desta, sua utilização no diagnóstico ainda parece limitada 17 .…”
