Heterogeneity in Subcutaneous Adipose Tissue Morphology and Metabolic Complications in Overweight and Obese Women

Abstract: In overweight/obese premenopausal women, larger adipocyte diameter in s.c. abdominal, but not gluteal, AT associates with low plasma adiponectin and systemic insulin resistance, and suggests that increased collagen thickness (obesity-related scarring) could contribute to these findings.

“…Many reports have shown a negative association between SC FCS and adiponectin release 19,61,69,103,105,106 , even after adjustment for SC adipose tissue area 19 . One study reported no association between OM FCS or SC FCS and adiponectin levels 24 .…”

“…Adipocyte cell size correlates positively with secretion of proinflammatory adipocytokines, e.g., leptin, inteleukin 6 and 8 (IL-6, IL-8), and monocyte chemoattractant protein-1 (MCP-1), as shown by data from cultured adipocytes [ 81 ]. In humans, visceral adipocyte size correlates directly with leptin [ 45 ] and inversely with adiponectin [ 82 ]. Adipose tissue expansion is regulated by storage-related genes like DGAT2, SREBP1c and cell death activator (CIDEA).…”

“… 64 Furthermore, the fraction of DPP4+ fibroblasts (FAC3) 56 , 57 , 58 is increased with AT hypertrophy, which supports the finding that adipocyte hypertrophy is associated with the thickening of pericellular collagen. 101 In addition, trajectory inference and RNA velocity analyses also revealed that FAC2 fibroadipogenic progenitors in AT hypertrophy are predicted to become FAC fibroblasts instead of FAC1 preadipocytes in AT hyperplasia, thus highlighting the potential role of FAC2 programming in mature adipocyte morphology and AT fibrosis. Because increased pericellular collagen deposition may hinder adipocyte expansion, increased number of fibroblasts may also contribute to ectopic fat accumulation and insulin resistance by limiting further fat storage expansion in hypertrophied subcutaneous adipocytes.…”
