Heterocyclic N-Oxides - An Emerging Class of Therapeutic Agents

Abstract: Heterocyclic N-oxides have emerged as potent compounds with anticancer, antibacterial, antihypertensive, antiparasitic, anti-HIV, anti-inflammatory, herbicidal, neuroprotective, and procognitive activities. The N-oxide motif has been successfully employed in a number of recent drug development projects. This review surveys the emergence of this scaffold in the mainstream medicinal chemistry with a focus on the discovery of the heterocyclic N-oxide drugs, N-oxide-specific mechanisms of action, drug-receptor int… Show more

“…We then choose to test a subclass of nitroxy compounds for their capacity to inhibit this interaction. Nitroxy compounds have recently emerged as a privileged scaffold for applications in drug discovery 15, 16 . In particular, 1,2-oxazines represent a new structural framework with significant potential for structure diversification.…”

“…[126] The NÀOd ipole of various heterocyclic N-oxides can act as an itric oxide (NO) agonist. They are found either as core units in compoundso fbiological interest, or they serve as versatile synthetic intermediates.N -Oxidation entered mainstream drug discovery,c redited with an improvement of properties and therapeutic outcome.…”

“…[2] In fact, the chemistry and applications of aromatic N-oxides have been receiving increased attention over the last two decades due to their usefulness as potential anticancer, antibacterial, antihypertensive, antiparasitic, anti-HIV, anti-inflammatory, herbicidal, neuro-protective, and procognitive agents. [3,4] ArNÀOX are useful as protecting groups, auxiliary agents, oxidants, ligands in metal complexes, and catalysts. [5] Also, ArNÀOX have been shown to be useful in energetic materials applications such as propellants, explosives, and pyrotechnics.…”
