HES6 reverses nuclear reprogramming of insulin-producing cells following cell fusion

Abstract: To examine the mechanism by which growth-stimulated pancreatic β-cells dedifferentiate, somatic cell fusions were performed between MIN6, a highly differentiated mouse insulinoma, and βlox5, a cell line derived from human β-cells which progressively dedifferentiated in culture. MIN6/βlox5 somatic cells hybrids underwent silencing of insulin expression and a marked decline in PDX1, NeuroD, and MafA, indicating that βlox5 expresses a dominant trans-acting factor(s) that represses β-cell differentiation. Expressi… Show more

“…However, pancreatic progenitors are known to have been already specified at the 6-somite stage [52] , [53] and our results suggests that c Hes6-1 is an early marker for these cells, before Pdx1 . This agrees with the findings that mouse Hes6 is also expressed in pancreatic cells and that its overexpression causes up-regulation of Pdx1 expression [54] .…”

“…GFP-positive cells, indicating expression from the human insulin promoter-GFP transgene introduced by lentivirus-mediated gene transfer, were visualized by fluorescence microscopy after two days. Cells were then harvested for RNA isolation and determination of insulin mRNA level by quantitative RT-PCR 30 . Palmitate induced a dose-dependent decrease in the number of GFP-positive cells.…”
