Varicella-zoster virus (VZV) is a neurotropic alphaherpesvirus that causes chickenpox andshingles. ORF7 is an important virulence determinant of VZV in both human skin and nerve tissues, however, its specific function and involved molecular mechanism in VZV pathogenesis remain largely elusive. Previous yeast two-hybrid studies on intraviral protein-protein interaction network in herpesviruses have revealed that VZV ORF7 may interact with ORF53, which is a virtually unstudied but essential viral protein. The aim of this study is to identify and characterize VZV ORF53, and to investigate its relationship with ORF7. For this purpose, we prepared monoclonal antibodies against ORF53 and, for the first time, characterized it as a ~40 kDa viral protein predominantly localizing to the trans-Golgi network of the infected host cell. Next, we further confirmed the interaction between ORF7 and ORF53 by co-immunoprecipitation and co-localization studies in both plasmid-transfected and VZV-infected cells. Moreover, interestingly, we found that ORF53 lost its trans-Golgi network localization and became dispersed in the cytoplasm of host cells infected with an ORF7-deleted recombinant VZV, and thus ORF7 seems to play a role in normal subcellular localization of ORF53. Collectively, these results suggested that ORF7 and ORF53 may function as a complex during infection, which may be implicated in VZV pathogenesis. A strategy for zooming in and out the topological environment of a node in a complex network is developed. This approach is applied here to generalize the subgraph centrality of nodes in complex networks. In this case the zooming in strategy is based on the use of some known matrix functions which allow focusing locally on the environment of a node. When a zooming out strategy is applied new matrix functions are introduced, which give a more global picture of the topological surrounds of a node. These indices permit a modulation of the scales at which the environment of a node influences its centrality. We apply them to the study of 10 protein-protein interaction (PPI) networks. We illustrate the similarities and differences between the generalized subgraph centrality indices as well as among them and some classical centrality measures. We show here that the use of centrality indices based on the zooming in strategy identifies a larger number of essential proteins in the yeast PPI network than any of the other centrality measures studied.

We study a Gaussian matrix function of the adjacency matrix of artificial and real-world networks. In particular, we study the Gaussian Estrada index-an index characterizing the importance of eigenvalues close to zero. This index accounts for the information contained in the eigenvalues close to zero in the spectra of networks. Here we obtain bounds for this index in simple graphs, proving that it reaches its maximum for star graphs followed by complete bipartite graphs. We also obtain formulas for the Estrada Gaussian index of Erdős-Rï¿oenyi random graphs as well as for the Barabï¿oesi-Albert graphs. We also show that in real-world networks this index is related to the existence of important structural patterns, such as complete bipartite subgraphs (bicliques). Such bicliques appear naturally in many real-world networks as a consequence of the evolutionary processes giving rise to them. In general, the Gaussian matrix function of the adjacency matrix of networks characterizes important structural information not described in previously used matrix functions of graphs. 2The spectrum of a network-the set of its eigenvalues-provides important information about the structural and dynamical properties of the corresponding system. Most of the functions used to study network spectra give more weight to the largest modular eigenvalues. Then, the information contained in the eigenvalues close to the centre of the spectra, i.e, those close to zero, has remained totally unexplored in the study of graph spectra. Here we study a Gaussian matrix function that gives more weights to the eigenvalues closest to the centre of the spectrum of a network. Using this function we extract important structural information hidden in the spectra of networks, such as emergence of complete bipartite subgraphs (bicliques) which appear naturally in many real-world networks as a consequence of the evolutionary processes giving rise to them. These bicliques are also ubiquitous in random networks generated by preferential attachment mechanisms, such as the Barabï¿oesi-Albert model. In this work we provide a series of analytical results that pave the way for further analysis and uses of this Gaussian matrix function to understand network structure and dynamics.3
