Herpes Simplex Virus Type 2 Vaccines: New Ground for Optimism?

Abstract: The development of effective prophylactic and therapeutic vaccines against genital herpes has proven problematic. Difficulties are associated with the complexity of the virus life cycle (latency) and our relatively poor understanding of the mechanism of immune control of primary and recurrent disease. The types of effector cells and the mechanisms responsible for their activation and regulation are particularly important. Studies from my and other laboratories have shown that recurrent disease is prevented by … Show more

“…The virus travels via retrograde axonal transport and gains access to neuronal cell bodies within the sensory ganglia, thus establishing a lifelong, persistent infection (3,10,19,23,24,58). Reactivation from latency can occur during times of emotional or physical stress and can cause recurrent disease (3,24). During periods of reactivation, the virus is shed from the infected host, sometimes in the absence of clinical symptoms, and thus may have an increased chance of infecting additional susceptible hosts (20,24).…”

“…Vaccines that prevent herpes simplex virus infection may not only decrease acquisition but also disease severity, neonatal herpes, and the transmission of HIV. 82,83 Cytomegalovirus has also been the target of vaccinedevelopment efforts. Cytomegalovirus is the most common intrauterine infection in the United States.…”
