Heregulin and HER3 are prognostic biomarkers in oropharyngeal squamous cell carcinoma

Abstract: BACKGROUND Although heregulin and HER3 are frequently expressed at high levels in head and neck cancer, their prognostic value remains unclear. We explored the prognostic significance of heregulin/HER3 expression in patients with oropharyngeal squamous cell carcinoma (OPSCC), taking into account other HER family members as well as p16 status. METHODS Ninety-six primary tumor specimens from OPSCC patients were retrospectively collected and analyzed for heregulin mRNA by in situ hybridization and for HER3, EGF… Show more

“…Exclusion of the outlying study did not change the results significantly (OR for 5 year OS: 1.30, 95% CI 0.78–2.14, p = 0.31). Similar results were observed in the two studies [24, 32] that reported HR for OS (pooled HR: 3.47, 95% CI 0.78–15.47, p = 0.10, Figure 2B) and when OS was examined at 3 years (pooled OR: 1.25, 95% CI 0.80–1.95, p = 0.33).…”

“…Once again, there was significant heterogeneity (Cochran Q p < 0.001, I 2 = 82%), although for PFS caused by general heterogeneity and not individual outlying studies. Similar results were observed in the two studies [24, 32] that reported HR for PFS (pooled HR: 1.64, 95% CI 0.94–2.86, p = 0.08, Figure 3B) and when PFS was examined at 3 years (pooled OR: 2.17, 95% CI 0.76–6.21, p = 0.15).…”

“…Several reports examining large cohorts of HNSCC patients have found HER3 expression to be associated with decreased survival, however, these early studies did not include HPV status in their evaluations (44). More recently, one group reported that HER3 and neuregulin 1 (NRG1), the cognate ligand for HER3, are independently associated with decreased overall survival in oropharyngeal cancer patients (45). In the current study, HER3 was overexpressed in HPV(+) HNSCC cell lines, PDX models, and pharyngeal tumors, where high levels of HER3 protein expression was associated with reduced survival of pharyngeal cancer patients.…”

“…They also found that patritumab, an antibody specific for HER3, is able to overcome such resistance. Our recent research identified that high heregulin mRNA and high HER3 protein levels are independent prognostic factors for poor overall survival in patients with oropharyngeal squamous cell carcinoma (OPSCC) (38). Our current study further demonstrated both in vitro and in vivo that inhibiting HER3 could resensitize cetuximab-resistant HNSCC cells to this agent.…”
