Hepatoma-selective imaging of heavy metal ions using a ‘clicked’ galactosylrhodamine probe

Abstract: This study depicts the 'click' construction of a water-soluble galactosyl rhodamine that can selectively probe mercury ions internalized by hepatoma cells over other cancer cells.

“…Additionally, devices such as microfluidic chips can allow for the cycling of serum for accumulation of trace biomarkers such as circulating tumor cells (CTCs). For in vivo theranostics, the addition of a targeting agent (such as a glycoligand for a receptor expressed on specific cells and tissues) might favor the target‐specific, low‐side‐effect imaging and treatment of diseases.…”

“…To test the biospecificity of the receptor‐targeting imaging, Hep‐G2 cell line with a largely reduced ASGPr expression level (sh‐ASGPr) by transfecting the cells with an si‐RNA plasmid, and a panel of other cells with minimal ASGPr expression were used . As shown in Figure a,b, the DK1 ‐based glycosheet showed an obvious imaging effect only for raw Hep‐G2 cells rather than other control cells.…”

“…we also used an established cell line with largely reduced ASGPr expression level (sh-ASGPr) [27][28][29][30]. To our delight, reduction of the receptor expression led to a largely decreased impedance increment (Fig.…”

“…Hep-G2 cells with the expression of asialoglycoprotein receptors (ASGPr) that recognize selectively galactose [27][28][29][30] were incubated with the electrodes at two different cell concentrations (500,000 and 10,000 cells mL -1 ). As a result, we observed an evident R ct increase in the presence of Hep-G2 in a concentration-dependent manner ( Fig.…”
