Hepatocellular Carcinoma cells: activity of Amygdalin and Sorafenib in Targeting AMPK /mTOR and BCL-2 for anti-angiogenesis and apoptosis cell death

Tarek El-Sewedy,
Afrah Fatthi Salama,
Amro E. Mohamed
et al.

Abstract: Background Sorafenib (Sor) is the only approved multikinase inhibitor indicated for the treatment of HCC. Previous studies have shown that amygdalin (Amy) possesses anticancer activities against several cancer cell lines; we suggested that these compounds might disrupt AMPK/mTOR and BCL-2. Therefore, the current study used integrated in vitro and in silico approaches to figure out Amy and Sor’s possible synergistic activity in targeting AMPK/mTOR and BCL-2 for anti-angiogenesis and apoptosis ce… Show more

“…HCC development is strictly related to the overexpression of many proteins that play critical roles in hindering the apoptotic cascade, such as Bcl-2, Bcl-xL, and Mcl-1. SRF has been demonstrated to induce apoptosis by downregulating the Bcl-2 family proteins in the HepG2 cells . Another study also demonstrated that the occurrence of nuclear condensation and fragmentation in HepG2 cells is directly related to the dose of administered SRF .…”

“…Bcl-2 is for both survival and induced cell death, can lead to cancer growth and prevent cancer treatment. Bax is one member of the B-cell lymphoma (Bcl-2) family, belonging to pro-apoptotic protein and a major pro-apoptotic protein in the Bcl-2 family (El-Sewedy et al, 2023). It is activated by some stimuli from inner cells such as DNA damage, and cytotoxicity.…”

“…These results align with the upregulation of AMPK, HMGB1, beclin-1, and ATG5 mRNA levels, as well as the downregulation of mTOR and Bcl-2 levels. Unlike sorafenib, amygdalin can increase GSH level, reduce MDA level, and exhibit strong DPPH free radical scavenging ability ( El-Sewedy et al, 2023 ). These findings suggest that amygdalin holds significant potential for the treatment of hepatocellular carcinoma.…”
