Hepatitis B Virus Mutants with Precore-Region Defects in Two Babies with Fulminant Hepatitis and Their Mothers Positive for Antibody to Hepatitis B e Antigen

Abstract: ABSTRACT. Clones of hepatitis B virus (HBVIndividuals infected with HBV have HBsAg, representing the viral envelope, in the circulation. In addition, they have either HBeAg, which is closely related to the viral nucleocapsid (I), or anti-HBe. The presence of HBeAg or anti-HBe in the serum is significant for the immunopathologic status of hosts and also for the biology of HBV infecting them (2).Perinatal HBV infection, resulting in a persistent camer state of babies, is transmitted by camer mothers who are posi… Show more

“…7 The reduced chronicity rate of our cWHV8pcmP1 inoculum in neonates (Table 7) may reflect the inability of the infecting virus to produce e antigen, 7 which is believed to act as an immunotolerogen in the neonate. 19,20 Thus, analysis of the uncloned WHV8P1 inoculum for precore mutants and coinfection studies using mixed cWHV8P1 and cWHV8pcmP1 could further address these issues.…”

“…The e antigen of HBV has been under study for several years for its potential role as an immunotolerogen 19 and also in association with the natural selection and transmission of e-minus mutants of HBV, some of which may exhibit increased pathogenicity in different settings. 20 Although naturally occurring precore stop codon mutations have been identified among mixed field strains of WHV, 21 the patterns of e-antigen/ antibody seroconversion in WHV infection have not been well studied because the relevant assays have not been developed. Nevertheless, model studies using a genetically engineered e-minus mutant of WHV could help to address some of these issues.…”

“…4 Perinatal transmission of HBV by eantigen positive mothers correlates with a high incidence of chronicity in the offspring. 5,6 Furthermore, in adults with chronic HBV infection, e-antigen positivity has been linked to the elimination of e-antigen and core-antigen specific T-cells thus favoring viral persistence. 7 While the role of e-antigen in regulating the immune system is only partially understood, 8,9 its high evolutionary conservation in the Hepadnaviridae suggests the importance of its function.…”
