Heparan sulfate fine‐tunes stromal‐epithelial communication in the prostate gland

Abstract: Several studies reported the concerted and mutual communication between the prostate epithelium and stroma, which determines the final organ architecture and function, but gets awry in cancer. Deciphering the mechanisms involved in this communication is crucial to find new therapeutic strategies. HS sequesters a number of secreted growth factors and cytokines, controlling their bioavailability to the target cells, suggesting that HS is an important regulator of the extracellular matrix (ECM) and a key player i… Show more

“…The affinity of HS for FGFs is specified by the pattern and level of sulfation of the HS chains (Barbosa et al, 2021; Hassan et al, 2021; Xu & Esko, 2014). Several recent studies have probed the mechanism of HS regulation of FGF signaling by manipulating HS modifying enzymes or identifying functional domains of the HS chain.…”

“…It is beyond the scope of this review to cover the complexities of the fine structure of HS other than to point out the extremely large number of ligands that can interact with HS, which is evident in the entries listed in Table 4. Many excellent reviews have appeared on HS, and the interested reader is directed to these for further information on the diverse roles of HS in various cell and tissue contexts [20,261,[325][326][327][328][329][330][331][332][333][334][335][336]. “…In prostate cancer, the expression of syndecan-1 in epithelial cells decreases when cells are transformed and acquire invasive properties. This decreased expression is associated with a bad prognosis (37)(38)(39).…”
